Does it affect gameplay like how putting safety’s in a lb position will make you susceptible to a run play?
I don't believe there's any penalty for this. Feel free to run OLBs at DE

It won't affect anything other than your player's overall may go down. That won't affect the actual gameplay though, just a visual thing.

I have Lawrence Taylor and Derrick Thomas at RRE and RLE, as well as the regular left and right DE positions. LT is flexible, he can get secondary position for any/all of these options, but DT is limited to MLB or either (L/R) OLB spot. I'm not running Campus Heroes theme team, so i don't really pay any attention to this, i just use what i consider to be the "best" (available to me) players as edge rushers, which are clearly LT & DT.

I run a 4-3 and like to have the better coverage lbs up top so I was subing tj watt and LT at the end positions but noticed that it shows their overall dropping to well below my Grahm and long I have at DE. So I’m just not sure if it makes me liable against runs or not

Ahhh ok so anytime you sub say a saftey at a corner they still play with their good stats even though it shows overall drop too

Yes, that is absolutely correct. The player / card's OVR rating is essentially meaningless. I really wish they'd adjust this, it's super confusing to many players, especially those of us who just started or have only been playing a year or two in the modern era.
For example, the 91 OVR Mike Evans MCS card remains one of the premier safeties in the entire game. Abilities are everything in this game, more than stats, more than OVR rating. Speed kills, but generally speaking, the rest of the stats simply need to hit certain thresholds to be viable, but this can mean anything from 85-90 for many things, with 95+ being considered "elite" I guess. As with many things, it depends, but don't let that in-game number changing from position to position scare you away.
Cornerbacks are mostly terrible this year, might behoove you to consider running Safeties at one or both outside CB positions. Good luck, have fun! Hope my rambling makes a little sense.

Correct. The individual ratings remain the same, it's just a visual thing with the overall.
Now safeties at linebacker do take a hit in the running game but that's the only true drop I'm aware of.
